Adaptogenic Efficacy of Withania Somnifera (Ashwagandha) on Cortisol Regulation

Abstract

Withania somnifera (Ashwagandha) is a premier adaptogenic herb in Ayurvedic medicine, used for over 3,000 years to improve stress resistance and overall vitality. This double-blind, placebo-controlled study examined the effects of a high-concentration, full-spectrum Ashwagandha root extract (KSM-66) on stress, cortisol levels, and quality of life in 64 adults with a history of chronic stress.

Methods

Participants were randomized into two groups: the treatment group received 300 mg of KSM-66 Ashwagandha root extract twice daily for 60 days, while the control group received an identical placebo. Stress levels were assessed using the Perceived Stress Scale (PSS), and serum cortisol levels were measured at baseline and at the end of the study.

Results

The treatment group demonstrated a significant reduction in serum cortisol levels (27.9% reduction, p < 0.0001) compared to the placebo group (7.9%). Scores on the PSS were also significantly lower in the Ashwagandha group, indicating a substantial improvement in perceived stress resistance.

Key Findings

  • Cortisol Reduction: 27.9% average decrease in serum cortisol
  • Stress Score Improvement: 44% reduction in PSS scores
  • Sleep Quality: 72% of participants reported improved sleep onset latency
  • Safety Profile: No significant adverse events reported

Discussion

These findings support the traditional Ayurvedic use of Ashwagandha as a Rasayana (rejuvenative tonic) and validate its role in modern stress management protocols. The mechanism of action is believed to involve modulation of the hypothalamic-pituitary-adrenal (HPA) axis.

Conclusion

High-concentration Ashwagandha root extract safely and effectively improves an individual's resistance towards stress and thereby improves self-assessed quality of life. AharvaPharma's Ashwagandha Elixir uses the same standardized KSM-66 extract at clinical dosages.
